Well, manual sorting after using Boss or Loot, as they never get it spot on. This is partly because two mods I always used get borked by a mod manager and also I learned more by manual sorting.

On the other hand, mods at the bottom of the list are loaded last. Some mods will say they require to be the highest mod, or at the top of the load order. They are considered higher in the mod load order. Mods at the top of the list are loaded first.
